New FISH approaches to investigate Wolbachia persistance and regulation within Armadillidium vulgare

Thanks to fluorescent molecular probes, Fluorescence in situ hybridization (FISH) combines molecular identification, enumeration and localization of environmental bacteria. Wolbachia distribution in A. vulgare remains poorly characterized, though we know it is heterogenous. Large scale FISH will allow to map the endosymbionts within the host.

Alternatively the fluorescent probes can be directed against transcripts, in which case FISH allows to quantify gene expression. On the one hand, we can combine in the same experiment both FISH approaches (targetting bacteria/targetting transcripts) to correlate host gene expression with Wolbachia numbers, in single cells but also in tissues or organs. On the other hand, we can target both host and Wolbachia transcripts, to show how A. vulgare and Wolbachia influence each other through gene expression.

This work relies on genes of interest selected during the analysis of molecular banks produced in the projects EuWol and EndoSymbArt.
